描述: Catalyzes the transport of the major hydrophobic bile salts, such as taurine and glycine-conjugated cholic acid across the canalicular membrane of hepatocytes in an ATP-dependent manner, therefore participates in hepatic bile acid homeostasis and consequently to lipid homeostasis through regulation of biliary lipid secretion in a bile salts dependent manner (PubMed:11172067, PubMed:14570929, PubMed:19228692, PubMed:22619174, PubMed:23764895). Transports taurine-conjugated bile salts more rapidly than glycine-conjugated bile salts (By similarity). 活性测试: ACTIVITY REGULATION: The uptake of taurocholate is inhibited by taurolithocholate sulfate with an IC(50) of 9 uM. Pravastatin competitively inhibits the transport of taurocholic acid. Cyclosporin A, glibenclamide, rifampicin and troglitazonestrongly competitively inhibit the transport activity of taurocholate (By similarity). The canalicular transport activity of taurocholate is strongly dependent on canalicular membrane cholesterol content (By similarity) (PubMed:19228692). The uptake of taurocholate is increased by short- and medium-chain fatty acids. 复溶: Centrifuge the vial before opening. Reconstitute to a concentration of 0.1-0.5 mg/mL in sterile distilled water. Avoid vortex or vigorously pipetting the protein. For long term storage, it is recommended to add a carrier protein or stablizer (e.g. 0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose), and aliquot the reconstituted protein solution to minimize free-thaw cycles.
